Base-substitution changes involve the replacement of specific nucleotide bases by other bases which lead to the production of a messenger RNA molecule which contains an informational sequence which deviates from the normal or wild type. Frameshift changes involve the removal or insertion of bases in the DNA which results in changes in the "reading frame" of the genetic information at the level of protein synthesis. These types of changes are illustrated in figure 1. Such changes in nucleotide sequence may lead to the production of proteins containing an abnormal sequence of amino acids (missense) or to the production of a non-coding triplet which leads to chain termination and the production of a shortened protein (nonsense).
